Born in Medellín, Colombia, Juana Lotero López holds degrees in Economics from the University of Antioquia (2013) and Film and Television Directing from the National University of Colombia (2018). From 2021–2024, she specialized in Fiction Directing at the International Film and Television School (EICTV) in Cuba. She is the co-founder of Noctámbulos Cine, an independent film production company.

Her directing credits include the short films FRAGMENTS TO MASTER SILENCE and AMA (LOVE), both made during her time at EICTV. She also wrote, directed, and co-produced WILD STRAWBERRIES (2022), which premiered at the 61st Cartagena International Film Festival (FICCI), and SWEET DREAMS (2020), which premiered at the Huelva Ibero-American Film Festival and received a FACIUNI grant as well as the Best Fantasy Short Film Award at the Colombian Women’s Film Festival.

Her most recent short film, NIÑA CHILAPA (2025), won the India Catalina Award for Best Colombian Short Film at FICCI 64 and was selected for Volume 6 of Proimágenes Colombia’s Refresh catalog (2024–2025).

In addition to her filmmaking work, she served as a teaching assistant at the National University of Colombia and has participated in numerous workshops, labs, and training programs with filmmakers and artists, including Andrés Denegri, Paz Encina, Stephen Bayly, Pedro Freire, Basel Ramsis, Agustina Pérez, Irina Raffo, Rafael Ramírez, and Mariano Llinás.
